    abstract:
    In order to investigate the role of oxidative stress in the development of keratoconus, human keratoconic corneal fibroblasts(hKCFs.) were cultured and subjected to 100 μmol/L H2O2 in vitro. The expression of gene of antioxidant enzymes, inflammatory cytokines, MMPs, TIMPs, Collagen I and LOXLs was determined by real-time fluorescent quantitative PCR. Our results show that the expression of SOD-2, HO-1, NQO-1, ICAM-1, IL-1β/-6/-8 and MMP-1/-3 was significantly up-regulated, and the the expression of Collagen Ⅰ-α2, TIMP-1/-2/-3, LOXL-1/-3/-4 was down-regulated in hKCFs under oxidative stress. This finding suggests that oxidative stress may lead to the release of more inflammatory factors from hKCFs, imbalanced expression of MMPs and TIMPs, collagen degradation or cross-linking disorders, thus causing corneal stroma thinning and promoting the occurrence and development of keratoconus.
